What Is a Centrifuge Tube?

A centrifuge tube is a vessel engineered to withstand the radial and axial stresses generated during high-speed centrifugation without deforming, cracking, or leaking. Conical-bottom tubes concentrate pelleted material at a single point for easy aspiration, while round-bottom tubes distribute stress more evenly around the base and are preferred for higher-RCF fixed-angle rotor applications where a sharp conical point would create a stress riser.

Material selection drives chemical compatibility and maximum g-force rating: polypropylene (PP) resists most solvents and autoclaving up to 121°C but tops out at lower RCF than polycarbonate (PC) or thick-wall polyallomer, which are rated for ultracentrifugation. Cap style — snap, screw, or o-ring sealed — determines whether a tube is rated for aerosol-tight biosafety use, a requirement in any protocol involving infectious or hazardous samples.

What's the difference between conical and round-bottom centrifuge tubes?

Conical-bottom tubes taper to a point, concentrating pelleted material for easy aspiration with a pipette — the standard choice for cell pelleting and general sample prep. Round-bottom tubes distribute mechanical stress more evenly around a curved base, which makes them better suited to higher-RCF fixed-angle rotor applications where a sharp conical point would otherwise create a stress concentration point during a run.

How do I know the maximum RCF a tube can handle?

Every centrifuge tube is manufacturer-rated for a maximum relative centrifugal force (RCF, expressed in x g) at a specified wall thickness and temperature. This rating accounts for the tube's material, wall thickness, and geometry — running a tube beyond its rating risks cracking, deformation, or catastrophic failure that can also damage the rotor and centrifuge. Always check the rating against your protocol's spin speed and rotor radius, not just the raw RPM.

Why does tube material (polypropylene vs. polycarbonate vs. polyallomer) matter?

Polypropylene is the most chemically resistant and autoclavable up to 121°C, but has a lower maximum RCF than polycarbonate or polyallomer. Polycarbonate offers higher clarity and higher-speed ratings but is attacked by many organic solvents. Polyallomer, a blend of polyethylene and polypropylene, is the standard for ultracentrifugation because it combines high chemical resistance with the mechanical strength needed at very high g-forces.

Do I need sterile or RNase/DNase-free tubes?

For molecular biology work — PCR, RNA extraction, cloning — RNase/DNase-free certified tubes are required to prevent sample degradation or contamination that would invalidate results. For cell culture, gamma-irradiated sterile tubes prevent microbial contamination of live cultures. General lab use (buffer prep, non-biological samples) can typically use non-certified bulk tubes at lower cost.

What cap style should I choose for hazardous or infectious samples?

O-ring sealed screw-cap tubes are required for aerosol-tight centrifugation of infectious, radioactive, or otherwise hazardous samples, since they prevent aerosolized material from escaping during high-speed spins or if a tube leaks. Standard snap-caps are adequate for routine, non-hazardous sample handling where aerosol containment isn't a biosafety requirement.
